Study Finds Origin Of Ovarian Cancer – A Knowledgeable Report

“Humans used cells from women with BRCA-1 mutations who had ovarian cancer to make these fallopian organoids,” said Clive Svendsen, PhD, executive director of the Cedars-Sinai Board of Governors Regenerative Medicine Institute.

The researchers used induced pluripotent stem cells (IPSCs), which can produce any type of cell, to achieve their discoveries.

They began by taking blood samples from two groups of women: young ovarian cancer patients with the BRCA-1 mutation and a healthy control group.

The iPSCs were then used to create organoids that mimicked the lining of fallopian tubes, and the organoids in the two groups were compared.

Nur Yucer, PhD, a project scientist in Svendsen’s group and the first author, remarked, “We were startled to identify many cellular abnormalities consistent with cancer development exclusively in the organoids from the BRCA-1 patients.”

Yucer continued, “Organoids generated from women with the most aggressive ovarian cancer revealed the most severe organoid disease.” The organoid method might be used to study how ovarian cancer is “seeded” in the fallopian tubes of women with mutant BRCA-1, as well as to assess if a treatment might work against the disease in an individual, according to Svendsen.

Each organoid was genetically identical to the person who donated blood, making it a “twin” of that person’s own fallopian tube linings. The organoids can be used to evaluate a variety of medications without exposing the patients.

“This research represents an exciting application of IPSCs, bringing us closer than ever to significantly improving the outcomes for women with this common type of ovarian cancer,” said Jeffrey Golden, MD, vice dean of Research and Graduate Education at Cedars-Sinai and director of the Burns and Allen Research Institute.

“Building on these discoveries, we may one day be able to provide early, lifesaving detection of ovarian cancer in women who possess the BRCA-1 mutation, as well as develop effective, customized prevention and treatment measures,” Golden noted.
